Molecular Data
Compound Overview
Thymosin Alpha-1 (Ta1) is a 28-amino acid peptide originally isolated from thymosin fraction 5, a thymus gland extract, by Dr. Allan Goldstein at George Washington University in the 1970s. It is one of the most clinically validated immune-modulating peptides, with regulatory approval in over 35 countries (as Zadaxin) for hepatitis B and as an immune adjuvant. Ta1 plays a central role in T-cell maturation and immune system homeostasis.
Reported Mechanism (Preclinical)
Thymosin Alpha-1 acts on toll-like receptors (TLR2, TLR9) on dendritic cells and innate immune cells, enhancing antigen presentation and stimulating both innate and adaptive immunity. It promotes T-cell differentiation from thymocyte precursors, enhances NK cell cytotoxicity, increases IL-2 and IFN-alpha production, and restores T-helper cell balance. It also activates indoleamine 2,3-dioxygenase (IDO), modulating immune tolerance and preventing excessive inflammatory responses.
Mechanisms described above are reported in preclinical (animal and in vitro) literature and have not been established for human use.
Key Research Highlights
- Improved survival in chronic hepatitis B patients when combined with interferon therapy (Andreone et al., 2001)
- Enhanced immune reconstitution in HIV-positive patients in clinical studies
- Showed efficacy as vaccine adjuvant, improving antibody response in elderly and immunocompromised populations
- Demonstrated anti-tumor immune effects by enhancing dendritic cell maturation and T-cell activation
- Improved outcomes in severe sepsis by restoring T-cell function and reducing inflammatory cytokine storms (Wu et al., 2013)
